Since this is usually the first question people ask, I wanted to start a discussion:
There's really three different types of combat: PVP Defense, PVP Offense and Campaign, so there's really no one "best team" for everything. I broke down the three scenario types and listed the best guys I could think of for each. You'll need to mix and match depending on what element types you are facing though, and a lot will also depend on how ascended and leveled your guys are. I would take a 5-star Zurk over a 1 star Torchy, even if Torchy is generally much better.
PvP Defense
This is really about preventing the big hits and trying to outlast your opponent.
Bovus - He used to be a must-have, but has been neutralized a bit with some of the updates. His Stampede attack gives -1 energy to the enemy, which means none of their special abilities will work on the first turn. That's crushing in PVP where most guys will die in the first 2 turns. He also gives a 15% HP increase to other Warriors on your team.
Yokozuma - His Sumo Stomp doesn't prevent attacks, but it really increases the chance that your enemy will miss on a lot of their big attacks. This effect lasts 2 turns, which can really save your guys if your enemy is whiffing on all their big attacks. Unfortunately, he doesn't have a really good power attack so he's not going to be doing a lot of the killing, but his 15% health for Warrior Allies makes him a great guy to pair with Bovus.
Igorok - A little hard to get, but if you do get him, his Polar Pounding is one of the deadliest attacks you'll see. Not only does it hit most guys for a big chunk of health, but there's a good chance it will freeze them and take them out for an entire turn.
Julius - Also a little hard to get, but his Barrel Toss is similar to Yoko's Sumo Stomp, but actually hits for pretty good damage too.
Tsume - Tsume is one of the best characters in the game. The problem with PvP defense is that he usually doesn't use his attacks when he's best able to use them. If you do use him, place him as one of your last characters, so he can hopefully take some damage before he uses his Soul Shred. I've had several battles where everyone on my team was eliminated, except for Tsume with a sliver of health, and he'd Soul Shred everyone to a one-hit instant death.
Chief NubNub - Although, I wouldn't really recommend a healer for PvP, if you get several stars on NubNub, I'd consider using him. If you have guys that aren't one shot kills, and he can get off a Voodoo Chant, it's game over. His Voodoo chant will heal through just about any attack your opponent can dish out. As long as you have other damage dealers alive, you will be a tough guy to finish off. He also is usually a priority target, so if he's 4-star rated or better, he can usually eat a few attack that your damage dealers would otherwise be taking.
Willow - She's great because if you can get her ascended, she's got three pretty good special attacks that you can use pretty much every turn. Added to that, she'll usually go first every turn and has a pretty good dodge ability, which keeps her alive. I'd just be wary of using her with 3 other green heroes, as someone could easily bring in an all Red team and tear right through you.
PvP Offense
Tsume - I can't count the number of times Tsume has saved my butt with a last second Soul Shred that wiped out a full strength opponent. The trick to Tsume is to save his best attacks until he's around half-health or worse. He's usually good for 3 or 4 decent sized attacks, so if you can time it right you can place him at the end of your lineup or skip a special attack in the first round, take a few hits of damage, and then rip through everyone the next turn.
Torchy - With the new update, Torchy is now one of the best offensive guys you can have. His first ascension gives you a bite that silences opponents. Since he's a fast attacker, that means you can nullify that Bovus Stampede that would otherwise render your team useless. He's also a fire type, so he can make quick work of your usual green-type defenders.
Lady Nim - She probably has the best single-shot attack in the game with "Get Over Here", which also Dazed opponents if it doesn't outright kill them. She's also your best option against Tsume, who you don't want hanging around with low health shredding through your team. Just make sure he's around 3/4's health or less and her shot should finish him off.
Bovus - Like in defense, his Stampede attack is critical in rendering the other team useless for a turn. If you can pair him with Torchy, you can block an other team's Stampede and then get off one of your own.
Campaign
A lot will depend on what elements you are facing on each map
NubNub - Nub is almost always a must-bring. Voodoo Chant last three turns and will keep you alive through just about everything. Even going up against all-fire teams, NubNub can usually stay alive just fine. I prefer his 3 turns of powerful healing with a refresh after 5 to Yasmin's one weak heal every 5 turns. The Team Defense and Haste can also helpful in boss fights.
